I would say that Sergey Karjakin is the best one out there now. He gets bad positions most of the time and somehow manages to draw it. 'Minister of Defense' is his other nickname.

Petrosian takes it for me. Many other players including Fisher have said he was nearly impossible to beat. Lev Polugaevsky even said it was easier to win a championship than to beat him

If the metric is percentage of games lost, Petar Trifunović has to be considered one of the greatest defensive players. He was known as a very difficult GM to beat, and lost only 9.3% of his games -- 65 losses in 697 games.
