If you play more positionally, and are more confident in the endgame, then I recommend the Berlin defense. If you like an agressive opening, play the Morphy defense. (Remember that this in my opinion, personal preferences still matter)
How to counter the Ruy Lopez? (Theo always plays that)

I found that most GMs prefer the Ruy Lopez Berlin Defense, hitting the pawn on e4, However, the Berlin and the Morphy is interchangable, see below: (The second one does not make sense)
The Stenitz Defense is also satisfactory, protecting the e5-pawn but letting the knight get pinned.
These three defenses are favoured by me, but it really depends on your style of playing. If you can't find a way to defend, just avoid it using Petroff's, Sicilian or Nimzowitsch. They will offer much better gameplay for black.
