there isn't a "best" opening
Best Opening

Well I was thinking more along the lines up getting my queen out in my first two moves, or setting my knights out close to the middle, which, if I'm not mistaken, is Knight's Defence?

Generally it's not a great idea to try to get your queen out before your other pieces, but if that's how you want to play, give it a shot. Learning from experience is a good thing too. ;)

1) Develop a central pawn to open lines for your pieces
2) Develop Knights before Bishops
3) Castle Early to Protect your King
4) Don't move a piece more than once in the opening if you can avoid it
5) Don't develop your Queen too early
5) Develop with a threat if you can
6) Develop the Queen to a safe square off of the first rank (somewhere where the opponent will have to go out of their way to attack it)
7) place your rooks on open files
These are the basics, pracitce these basics and you will be off to a better start than someone trying to memorize one specific opening.
In addition to all the guidelines that jwhitesj gave, try to pick openings that make it difficult for your opponent to do the same things. i.e., fight for the center to keep your opponent cramped, position your pieces so that they stop the opponent's king from castling (if it's convenient for you), force your opponent to move pieces twice so that he/she wastes time, don't let your opponent make too many threats, etc. Don't start memorizing too many opening lines until you really understand all of these principles.
Hey. What would be the best opening moves a person could make?
