No. 'No vacation' applies to tournaments only.
Only 6 parameters can be specified on individual challenges:
Starting position, days per move, colour, rated/unrated/takeback, game name & whether against a named individual or an open seek.
By contrast, there about 18 facets that can be controlled on tournament creation - hence the desirability of a tournament selection wizard to help filter the choices.
Is it possible to include a 'no vacation' feature when I put out a new game invite, either generally or to a specific player?
